Overview

IGF-1 DES (Des(1-3) IGF-1) is a naturally occurring truncated form of IGF-1 found in human brain tissue, missing the first three N-terminal amino acids (Gly-Pro-Glu). This truncation eliminates most binding to IGF binding proteins (IGFBPs), making it 5-10 times more potent than native IGF-1 at the receptor level. However, unlike IGF-1 LR3, it has a very short half-life of only 20-30 minutes, making it ideal for site-specific intramuscular injections targeting local muscle growth. IGF-1 DES is primarily used in bodybuilding for its powerful localized anabolic effects, including satellite cell activation and muscle hyperplasia. Its short half-life means it acts intensely but briefly, reducing systemic side effects compared to IGF-1 LR3.

Mechanism of Action

IGF-1 DES binds IGF-1 receptors with normal affinity but is virtually free from IGFBP sequestration: (1) Activates PI3K/Akt/mTOR for rapid protein synthesis at the injection site; (2) Potently stimulates satellite cell proliferation and differentiation for localized hyperplasia; (3) Short half-life concentrates effects at injection site before systemic clearance; (4) Enhances local glucose and amino acid uptake; (5) Promotes autocrine/paracrine signaling in injected muscle tissue; (6) Less systemic insulin-like effects than IGF-1 LR3 due to rapid clearance.
