Snap-8

Cosmetic Neuropeptidecosmetic

Also known as: Acetyl Octapeptide-3, SNAP-8 Peptide, Botox Alternative Peptide

Overview

Snap-8 (Acetyl Octapeptide-3) is an elongated version of Argireline (Acetyl Hexapeptide-3) with two additional amino acids, designed to more effectively inhibit SNARE complex assembly. The SNARE complex is essential for the fusion of synaptic vesicles with the presynaptic membrane, which releases acetylcholine at the neuromuscular junction. By competing with SNAP-25 (one of the three SNARE proteins), Snap-8 reduces muscle contraction intensity in facial muscles, thereby smoothing expression lines and wrinkles — similar to botulinum toxin but through topical application rather than injection. Studies suggest Snap-8 may be up to 30% more effective than Argireline at reducing wrinkle depth. It is widely used in cosmeceutical formulations and is one of the most popular anti-aging peptides in skincare.

Mechanism of Action

Snap-8 mimics the N-terminal domain of SNAP-25: (1) Competes with native SNAP-25 for incorporation into the SNARE complex; (2) Prevents proper assembly of the SNARE complex (syntaxin-1, SNAP-25, VAMP/synaptobrevin); (3) Reduces synaptic vesicle fusion and acetylcholine release; (4) Decreases the intensity of muscle contractions underlying expression wrinkles; (5) Does not paralyze muscles — reduces contraction force by approximately 30-40%; (6) Effect is localized to the application area and fully reversible upon discontinuation.
